Özet
ÖZET Bu araştırma, yurdumuzda yaygın olarak üretimi yapılan Agaricus bisporus' un spor çimlenmesi ve misel gelişmesi üzerine değişik ortamların etkileri saptanarak, misel kalitesi ve verimini artırmak amacıyla yapılmıştır. Denemeler 1994-1995 yılları arasında Ege Üniversitesi Ziraat Fakültesi Bahçe Bitkileri doku kültürü laboratuvarında gerçekleştirilmiştir. Çalışmada, minimal, Lambert, buğday-agar (BA), sentetik (St), patates-dekstroz-agar (PDA), malt-maya-agar, patates- dekstroz-agar-maya (PDAY), Yang, malt-agar, buğday-agar-maya (BAM) ortamlarını taşıyan ortam serisi kullanılmıştır. En hızlı çimlenme BAM (7gün), malt-maya-agar (7gün), sentetik (8gün) ortamlarında gerçekleşmiştir.Lambert <13gün) 2., PDA(15gün) 3., buğday-agar (BA) (23gün) 4., PDAY (48gün) 5., Yang (53gün) 6. sırada yer almıştır. Ortamlar petriyi tamamlama hızına göre son ölçüm değerleri baz alınarak sıralandığında sonuçlar 1. PDA (6364,29mm-), 2. Buğday - agar (6288,123mm-), 3. Yang (6034,93mm2), 4. Malt-maya- agar (6011, 033mm2), 5. Buğday - agar-maya (5773,361mm2), 6. PDAY (5625,861mm2), 7. Sentetik (5005,431mm2), 8. Lambert (3619,478m- m2), 9. Malt-agar (3154,09mm2), 10. Minimal (406,836mm2) olarak bulunmuştur. Lambert, Buğday-agar, Buğday-agar-maya mutlak olarak ışınsal ipliksi gelişmiş, diğerleriyse pamuksu veya karışık gelişmiştir (pamuksu+ ipliksi)
SUMMARY This research has been carried out in order to different nutrient media on germination of spore and growth of mycelium of Acraricus bisporus which is widely cultivated in our country. Experiments were developed at tissue culture laboratory of Ege University Agriculture Faculty department of Horticulture. Three of the fastest spore germinator media were found Wheat-agar-yeast (7days), Malt-yeast-agar (7days), Sentetic (8days). Lambert was 2. (13days), PDA was 3. (15days), Wheat-agar was 4. (48day), Yang was found 5. (53days). Any spor germination was not occured in Mal t -agar and Minimal media. Media were compared about the developing rate and 1. PDA (5364,29mm2), 2. Wheat-agar (6288,123mm-), 3. Yang (6034,93mm2), 4. Malt-yeast-agar (6011,033mm2), 5. Wheat -yeast -agar (5773,361mm2), 6. PDAY (5625,861mm2), 7. Sentetic (5005,431mm2), 8. Lambert (3619,478mm2), 9. Malt-agar (3154,09mm2), 10. Minimal (406,836mm2) were found. Lambert, wheat-agar, wheat -yeast -agar were absolutely developed strandy type and radial form. Others were developed flufy or mixed (flufy + strandy).
