Özet
ÖZET Diaminoksidazlar(amin:oksijen oksidoredüktaz, EC 1.4.3.6) bitki, hayvan ve mikroorganizmalarda yaygın olarak bulunan ve farklı tekniklerin kullanımıyla çeşitli kaynaklardan izolasyonu ve saflaştırılması üzerine çalışmalar yapılmış bir enzim grubudur. Bu grup enzimler,metabolizmada çeşitli semptomlara neden olabilen biyojen aminlerin; gıda ve fizyolojik sıvılardaki tayinlerinde kullanılabilir olmalarından dolayı, farklı kaynaklardan saflaştınlmalan önem kazanmaktadır. Bu çalışmada Diamin oksidaz kaynağı olarak, literatür verilerine göre Diaminoksidazca zengin olarak belirtilmiş yer fıstığı seçilmiştir. Çalışmada, saflaştırma amacıyla kullandığınız metod homojenizasyon, ultrafiltrasyon ve affinite kromatografisi adımlarını içermektedir. Bu çalışmada Histamin Diamin Oksidaz 1865 kat saflaştırıldı. îleri aşamada ise saflaştınlan enzimin karakterizasyonu yapıldı. Bulguların, bu konuda önceki çalışmalarla karşılaştırılması ve yorumu Sonuçlar ve Tartışma kısmında verildi. 41

SUMMARY Diamine oxidase(amine: oxygen oxidoreductase, EC 1.4.3.6), is widely distributed in animals, plants and microorganisms and it has been isolated and purified with different techniques from various sources. It is useful to determine the biogenic amine content of various foods, that has been widely studied because of their potential toxicitiy. For this reason.isolation and purification of DAO from different sources has a great importance. In this study Peanut were chosen as a source of DAO. Because the enzyme activity was high in comparison with the other species. For the purification of DAO, we describe a novel procedure which includes homogenization, ultrafiltration and affinity chromatography respectively. This technique resulted in 1865 fold purification with a considerable time-saving, compared to less selective multistage procedures. Besides, characterization of the purified enzyme was performed and compared with recent works. 40
