İn vitro testlerde başarılı bulunan 24 EB izolatı in vivo saksı denemeleri için seçilmiştir. EB'ler sisteme tohum kaplama ve kök içirme biçiminde iki kez uygulanmıştır. EB uygulaması gören bitkiler FOC spor süspansiyonu ile bulaşık torfa şaşırtılmıştır. Saksı denemelerinde, izolatların % 25'i, in vivo koşullar altında % 32 - 62 oranında hıyar bitkilerinde FOC'un gelişimini engellemiştir. 83 ve 99 no.lu EB'lerin bitki içinde zamana bağlı olarak kolonizasyonu periyodik olarak değerlendirildiğinde, FOC'un 83 no.lu bakterinin kolonizasyonu üzerine olumsuz etkisi saptanmazken, 99 no.lu EB popülasyonu FOC'un varlığından olumsuz etkilenmiştir. FOC'un bitki içindeki kolonizasyonu ise, EB uygulamalarından etkilenmemiştir. Bu sonuçlar, EB'lerin konukçu bitkide sistemik dayanıklılığı uyarma potansiyeli olabileceğini göstermiştir.
In this study, 109 EB strains were isolated from internal tissues of healthy cucumber plants which were grown in greenhouse and field in Turkey. These isolates were screened in vitro for their plant growth promoting traits and antagonistic activity against Fusarium oxysporum f. sp. cucumerinum (FOC). Out of 24 EB isolates exhibiting biocontrol and multiple plant growth promoting (PGP) traits were tested in growth chamber and greenhouse experiments. EB inoculation took place two times; before sowing as seed coating and after transplanting as substrate drenching. Cucumber plants with treated EB were transplanted to peat inoculated with FOC spore suspension (106 spor ml-1) in order to observe the suppression of Fusarial wilt on cucumber plants. In pot experiments, 25% of the EB isolates inhibited Fusarium wilt disease on cucumber plants at the rate of 32 to 62% compared to pathogen alone treatment. Out of the isolates, EB strain 83 (Pantoea agglomerans) and 99 (Bacillus thuringiensis) were selected for further greenhouse experiments. Acibenzolar-s-methyl (ASM) was also tested as reference chemical for comparing the efficacy of EB strains in terms of inducing the systemic resistance in cucumber plants. The treatments of EB strains 83 and 99 inhibited the disease severity of FOC at the rate of 53% and 49%, respectiveley, compared to pathogen alone treatment, and found as effective as ASM treatment. Moreover, EB strain 99, without being any disease pressure, increased the total marketable fruit yield at the rate of 30%, compared to non treated cucumber plants in soilless cultivation. It was also monitored the colonization and population dynamics of EB strains in the plant tissues for 75 days. The population density of EB strains in cucumber plants was determined as approximately 103-105 cfu plant g-1. We suggest that EB strains have the potential to activate inducible plant defense systems.
