Özet
Bu çalışmada, Türkiye'de yabani olarak yetişen bir Ranunculus türü olan Ranunculus constantinopolitanus DC. bitkisi farmakognozik açıdan incelenmiştir. Botanik çalışmalarda, bitki tayininde yararlanabilecek ve muhtemel monografilerde yer alabilecek anatomik ve mikroskobik özellikleri araştırılmıştır. Kalite kontrol çalışmasının yapıldığı araştırmalarda, çiçekli vejetasyon döneminde toplanan bitkiden elde edilen drog üzerinde Avrupa Farmakopesi'nde yer alan yöntemler esas alınmıştır. Bu amaçla, kurutmada kayıp, total kül, hidroklorik asitte çözünmeyen kül ve sülfat külü deneyleri yapılmış ve sonuçları % değer olarak verilmiştir. Ayrıca, droğumuzdan hazırladığımız ekstrelere fitokimyasal tarama yapılmıştır. Droğun biyolojik aktivitesinin belirlenmesi için çeşitli yöntemler gerçekleştirilmiştir. Droğun sitotoksik aktivitesinin belirlenmesi için, brine shrimp ve WST-1 hücre proliferasyonu yöntemleri yanında, antimikrobiyal aktivitesi disk difüzyon ve mikrodilüsyon yöntemleriyle; antioksidan aktivitesi DPPH radikali yakalama yöntemi ile belirlenmiştir. Ayrıca toplam fenolik madde analizi ile toplam flavonoit madde analizi de yapılmıştır.
In this study, Ranunculus constantinpolitanus DC. which is wild-growing Ranunculus species in Turkey, has been thoroughly investigated by pharmacognostical approaches. In botanical investigations carried out on the plant, anotamical and microscobical features, which may be used in a prospective monograph about this plant drug were investigated. In this course of the quality control studies have been conducted according to European Pharmacopeia, loss in drying, total ash, acid-soluable ash and sulphated ash of drug were determinated, and the results were report as percentage values. In addition, the phtochemical screening was investigated extracts prepared from our plant. To determinated the biological activity of the drug has been studied by using different methods. The cytotoxicity of the drug was evaluated by brine shrimp and WST-1 cell proliferation lethality bioassays; the antimicrobial activity of the drug was determinated by using disc diffusion method and microdilution method; the antioxidant activity of the drug was determinated by DPPH method. Moreover, analysis of total phenolic substance and total flavonoid substances were studied.
