Özet

In this thesis, microglia were differentiated from healthy induced pluripotent stem cells in order to model neuroinflammation associated with neurodegeneration in Rett syndrome (a rare neurological disorder caused by faulty genes, for which a very good biomimetic preclinical model has not been developed yet) and the effect of neuroinflammation on glial cells in the microenvironment. In the differentiation process, different times were studied for the transition to the maturation stage (late, middle, early stages), time optimization was made for high-yield differentiation, and it was determined that the best time was the middle stage with min. 84.4% yield. Then differentiated cells was characterized by FACS analysis, immunofluorescence and microscopic observation, RT-PCR analysis with particularly marked by maturation markers CD11b, CX3CR1, P2RY12, IBA1 etc. The effect of glutamate expression, which is secreted as an inflammatory response by differentiated and characterized microglia cells, lipopolysaccharide-activated, activated/non-activated microglia cells, on oligodendrocyte cells was evaluated by microscopic observation, immunofluorescence, microglial markers such as CD11b, CX3CR1, CD14, and oligodendrocyte markers such as MBP, GFAP, S100B, OLIG2 by RT-PCR on morphological and gene basis. The effects of glutamate on demyelination, progenitor cell proliferation and cell viability were revealed.
