16 farklı toprak örneğinden 139 Aktinomiset straini izole edilmiş, 104 strainin glukoz İzomeraz ürettiği saptanmış ve bunların arasından 1-8 nolu strainin en iyi GI üreticisi olarak belirlenmiştir. Bir Streptomyces sp. olarak tanımlanan bu strain, %1 ksiloz, %2 mısır ıslatma sıvısı, % 0.1 MgSC>4 7H2O ve % 0.01 C0CI2 6H2O içeren bir ortamda, 200 rpm* de, pH 7.0 de, 30 °C de, 24 saat büyütüldüğünde en yüksek glukoz izomeraz üretmiş; diğer taraftan 70 °C de, pH 7.0' da 1 saatlik inkübasyon sonunda en yüksek GI aktivitesi göstermiş; glukoz, Mg+2 ve Co+2 için Km değerleri 0.268 M, 60 mM ve 3.4 mM olarak saptanmıştır. 70 °C'de 84 saat yanlanma süresi olan bu enzimin oldukça termostabil olduğu görülmüştür. Actinomycetes strains were isolated from the different soils; 104 of the 139 strains showed glucose isomerase activity. As 1-8 numbered strain was determined best glucose isomerase producer among the 104 strains. 1-8, identified as a Streptomyces sp, grown in the medium contained %1 xylose, %2 cornsteep liqueur, %0,1 MgS04-7H20 and %0,01 C0CI2 6H20, at pH 7.0 at 30°C and at speed of 200 rpm for 24 hours showed best production of glucose isomerase; on the other hand, this strain showed best activity at pH 7.0, at 70°C for an hour; the michealis constant for glucose, Mg2+ and Co2+ were found 0.268 M, 60 mM and 3.4 mM, respectively.
