Özet
Tez üç bölümden oluşmaktadır. Başlangıçta Schiff bazlarının genel özellikleri sentez yöntemleri ve komplekslerin uygulama alanlarına ilişkin bilgiler ile Alzheimer hastalığı ve Alzheimer hastalığında kullanılan ilaçlar yer almaktadır. İkinci bölüm ise gereç ve yöntemi içermektedir. Çalışmada bir tanesi literatürde olan iki tanesi yeni 3 adet N-izopropiletilendiaminden türeyen Schiff bazı(I, II, III) sentezlenmiştir. Bu schiff bazları ile şelat yapılı Pt(II) kompleksleri (IK, IIK, IIIK) sentezlenerek yapıları elementel analiz, FT-IR, 1H-NMR, 13CNMR ile saptanmıştır .IK Kompleksinin yapısı X-ışınları Kırınım Yöntemi ile aydınlatılmıştır.Schiff bazlarının hem amin hem de imin azotundan metale bağlandıkları anlaşılmıştır. Komplekslerin genel yapısı aşağıda gösterilmiştir. IK için; R1=OMe;R2=OMe; R3=OMe, IIK için; R1=OMe;R2=OCH2Ph; R3=H, IIIK için; R1=H;R2=N(CH3)2 ; R3=H Sentezlenen bileşiklerin öncelikle Histidin ile etkileşimleri 1H- NMR ve kütle ölçümleri ile incelenmiştir. Daha sonra ise Aβ1-42 ile SHSY-5Y hücre hatları üzerindeki in vitro etkileşimleri saptanmıştır. Bunu takiben Tiyoflavin T ile floresans ölçümleri alınmıştır. Üçüncü bölüm ise Bulgular ve Tartışma kısmı olup, öncelikle Schiff bazlarının ve bunların Pt komplekslerin yapılarının çeşitli spektroskopik yöntemlerle aydınlatılması ve biyolojik çalışmalarının değerlendirilmesi ile ilgilidir. Komplekslerin lipofilite özellikleri elektronik spektroskopi yardımıyla belirlenmiştir. Bu komplekslerin Alzheimer tedavisindeki etkinliklerinin bir ön çalışması olarak histidin ile etkileşimleri 1H-NMR ile izlenmiş ve her üç kompleksinde gerek 1H-NMR gerek se LC/Q-TOF/MS sonuçları ile 1:2(Kompleks/Histidin) oranında etkileştiği saptanmıştır. Aβ1-42 ile etkileşimleri in vitro hücre çalışmaları ile ortaya konmuştur.
The thesis consists of three parts . The initial part consist of the properties of Schiff bases, synthesis methods and application areas of complexes, and Alzheimer's disease and drugs used in Alzheimer's disease are included. The second part includes materials and methods. In this study, Schiff base (I, II, III) was synthesized from three new N-isopropylethylenediamine, one of which is in the literature. These schiff bases and chelated Pt (II) complexes IK, IIK, IIIK) were synthesized and their structures were determined by elemental analysis, FTIR, 1H-NMR, 13C-NMR. The structure of IK complex was determined by the X-Ray differaction method. Schiff bases are understood to bind both from amine and imine nitrogen to metal. The general structure of the complexes is shown below. IK için; R1=OMe;R2=OMe; R3=OMe, IIK için; R1=OMe;R2=OCH2Ph; R3=H, IIIK için; R1=H;R2=N(CH3)2 ; R3=H The interactions of the synthesized compounds with Histidine were primarily investigated by 1H-NMR and mass measurements. Subsequently, in vitro interactions with Aβ1-42 SHSY-5Y cell lines were determined. Following this, fluorescence measurements were taken with Thioflavin T. The third part is the Results and Discussion section and it is primarily concerned with the elucidation of Schiff bases and their Pt complexes by various spectroscopic methods and evaluation of their biological studies. Lipophilic properties of complexes were determined by electronic spectroscopy. As a preliminary study of the efficacy of these complexes in Alzheimer's treatment, their interaction with histidine was monitored by 1 H-NMR and it was found that it interacts with 1: 2 (complex / histidine) ratio in all three complexes. In addition, the interaction of the complexes with LC / Q-TOF / MS with histidine was determined. According to the data obtained as a result of interactions with histidine. Their interactions with Aβ1-42 have been demonstrated by in vitro cell studies.
