Özet
Bu tez Nitron NHC (N-Heterosiklik Karben) komplekslerinin sentezi ve sitotoksik aktivitelerinin araştırılması ile ilgilidir. Günümüzde kanser tedavisinde sis-platin ve türevleri kullanılmaktadır. Fakat bu ilaçların toksik ve diğer yan etkilerinden dolayı yeni ilaç arayışları hızla artmaktadır. Platin metalinin yanında rutenyum ve iridyum içeren NHC kompleksler ile bu alanda umut verici sonuçlar elde edilmektedir. Nitron bir NHC ligantıdır ve bu metallerle organometalik bileşikler oluşturmaktadır. Bu nedenle bu çalışmada Nitron içeren platin, paladyum, iridyum, rodyum ve rutenyum kompleksleri sentezlenerek çeşitli kanserli hücre hatlarında sitotoksik aktiviteleri araştırılmıştır. Tez üç bölümden oluşmaktadır. İlk bölümde NHC bileşikleri ligand özellikleri, sentez yöntemleri ve Nitron benzeri mezo-iyonik karbenlerin sitotoksik aktiviteleri ile ilgili yapılmış olan bazı çalışmalara yer verilmiştir. İkinci bölümde deneysel çalışmalar açıklanmıştır. Üçüncü bölümde, sentezlenen komplekslerin karakterizasyonu ve sitotoksik özellikleri hakkında bilgi verilmiştir. Sentezlenen bileşiklerin reaksiyonları, Nitrondan yola çıkarak Ru, Rh, Pt, Pd ve Ir metal tuzlarının reaksiyonu sonucunda gerçekleştirilmiştir. Sentezlenen Ru-, Ir-, Pt-, Pd- ve Rh-Nitron kompleklerinin sitotoksik aktiviteleri MTT testleri ile incelendi. İnsan akciğer fibroblast (CCD-34Lu), insan meme epitelyal adenokarsinom (MDA-MB-231), insan prostat adenokarsinom (PC-3) ve insan serviks adenokarsinom (HeLa) hücre hatlarında incelendi. Sonuçlar göze alındığında kanserli hücreler üzerinde en iyi aktiviteyi [(Nitron)RuCl(p-simen)] kompleksinin verdiği anlaşıldı
This thesis is about synthesis of Nitron NHC (N-Heterocyclic Carbene) complexes and investigation of their cytotoxic activities. Today, cis-platinum and its derivatives are used in cancer treatment. However, due to the toxic and other side effects of these drugs, the search for new drugs is increasing rapidly. Promising results are obtained in this field with NHC complexes containing ruthenium and iridium as well as platinum metal. Nitron is also an NHC ligand and forms organometallic compounds with these metals. Therefore, in this study, platinum, palladium, iridium, rhodium and ruthenium complexes containing Nitron were synthesized and their cytotoxic activities were investigated in various cancer cell lines. The thesis consists of three parts. In the first part, some studies on the ligand properties of NHC compounds, synthesis methods and cytotoxic activities of nitron-like meso-ionic carbenes are given. In the second part, experimental studies are explained. In the third chapter, information about the characterization and cytotoxic properties of the synthesized complexes is given. The reactions of the synthesized compounds were carried out as a result of the reaction of Ru, Rh, Pt, Pd and Ir metal salts starting from Nitron. The cytotoxic activities of the synthesized Ru-, Ir-, Pt-, Pd- and Rh-Nitron complexes were examined by MTT tests. It was studied in human lung fibroblast (CCD-34Lu), human breast epithelial adenocarcinoma (MDA-MB-231), human prostate adenocarcinoma (PC-3) and human cervical adenocarcinoma (HeLa) cell lines. Considering the results, it was understood that [(Nitron)RuCl(p-cymene)] complex gave the best activity on cancer cells.
