Özet
Bu tezin amacı Pycnoporus cinnabarinus'tan fenoksazinon yapısındaki doğal bir pigment olan sinnabarinik asiti 3L'lik karıştırmalı tank reaktöründe üretip izole etmektir.Pycnoporus cinnabarinus tarafından 3L'lik karıştırmalı tank reaktöründe sinnabarinik asit üretimi için en uygun koşullar araştırıldı. Bu nedenle, çalkalamalı kültürde ve karıştırmalı tank reaktöründe en uygun üretim koşulları belirlendi. Yapılan optimizasyon denemeleri sonucunda; uygun karbon kaynağı 2,5 g/L maltoz, inkübasyon sıcaklığı 26ºC, aşılama miktarı %1 olarak bulundu. Kültivasyon, ortam pH'sında değişiklik yapılmadan gerçekleştirildi. Sinnabarinik asit en uygun koşullarda üretildikten sonra, ekstraksiyon işlemine tabi tutuldu ve TLC ve kolon kromatografisi kullanılarak ayrıldı. Spektrum taraması yapılan sinnabarinik asitin maksimum 450 nm'de karakteristik pik verdiği gözlendi.Kültür sıvısına turuncu renk veren ve antibakteriyel özelliği olan sinnabarinik asit farklı tekstil materyallerine uygulanarak alternatif kullanım alanı araştırıldı. Bu nedenle hem boyama özelliği hem de antibakteriyel etkisi araştırılan sinnabarinik asit için boyar madde endüstrisinde alternatif bir kaynak olarak yer bulacağı düşünülmektedir
The aim of this thesis was to produce the natural phenoxazinone pigment, cinnabarinic acid, from Pycnoporus cinnabarinus in 3L stirred tank reactor (STR) and isolate this pigment from the culture medium.The optimum conditions were investigated to produce cinnabarinic acid from Pycnoporus cinnabarinus in the 3L stirred tank reactor. Due to this cause, the optimum conditions were determined in the stirred tank reactor. As a result of optimization, the suitable carbon source, incubation temperature and inoculation rate were determined as 2,5 g/L maltose, 26ºC and 1%, respectively. All the cultivations were carried out at culture mediums pH. After the production of cinnabarinic acid under the optimum conditions, it was extracted. Afterwards these phases were separated by chromatography and determined with spectrophotometrically. The maximum absorbtion was observed in 450 nm.Cinnabarinic acid gives a orange color and an antibacterial property to the culture medium. Because of these effects an alternative usage in the textile industry for different textile materials were studied.
