Domates tohumlarında bazı viral etmenlerin bulunma durumlarının belirlenmesi ve güncellenmesi

Özet
Bu çalışmada ülkemiz ekonomisinde öneme sahip olan domates tohumlarında bulunan viral hastalık etmenlerin belirlenmesi, güncellenmesi ve daha önce domates tohumlarında bulunmadığı düşünülen ve test edilmeyen ancak son yıllarda yapılan araştırmalarda yeni ve hassas yöntemlerle bulunduğu belirlenen TYLCV ve PepMV gibi viral etmenlerin ülkemiz açısından kontrolünün gerçekleştirilmesi ve bu etmenlerin bulunmaları halinde gerekli kurumlarla sonuçların paylaşılarak test edilmesi gereken viral etmenlerin ve karantina listelerinin güncellenmesi amaçlanmıştır. Çalışma kapsamında 238 adet tohum örneği kullanılmıştır. Tohum örneklerindeki viral etmenlerin tanılanmasında serolojik ve moleküler yöntemler kullanılmıştır. Domates çeşitlerine ait 238 tohum örneği Tobacco mosaic tobamovirus (TMV), Tomato mosaic tobamovirus (ToMV), Cucumber mosaic cucumovirus (CMV), Tomato yellow leaf curl geminivirus (TYLCV), Tomato spotted wilt tospovirus (TSWV), Pepino mosaic potexvirus (PepMV), Pepper mild mottle tobamovirus (PMMoV) ve Tomato brown rugose fruit tobamovirus (ToBRFV) adlı etmenlerin varlığı açısından testlenmiştir ve sonuçta; örneklerin %12'sinin TMV, %23'ünün ToMV ve %8'inin CMV ile enfekteli olduğu görülmüştür. Araştırma kapsamında test edilen bazı tohum örneklerinde karışık enfeksiyonlar olduğu belirlenmiştir. Araştırmada toplam testlenen 238 domates bitkisi tohum örneklerinde 67 tane tohumda toplam enfeksiyon oranı % 28 olarak belirlenirken örneklerin en az bir adet viral etmenle bulaşık olduğu belirlenmiştir.
The aim of this study is to determine, update the viral disease agents in tomato seeds, which plays an important role for our country's economy, and to control, on behalf of our country, viral factors like TYLCV and PepMV, which were thought not to be existing in the past but were determined by new and sensitive methods in the recent years. It is also the aim, if viral agents like TYLCV and PepMV are found, to share this findings with the necessary institutions and to update the viral factors which needs to be tested, and the quarantine lists. 238 seed samples were used in this study. Serological and molecular methods were used in order to identify the viral agents in the seed samples. 238 seed samples of different tomato varieties were tested in order to prove the possible existence of Tobacco mosaic tobamovirus (TMV), Tomato mosaic tobamovirus (ToMV), Cucumber mosaic cucumovirus (CMV), Tomato yellow leaf curl geminivirus (TYLCV), Tomato spotted wilt tospovirus (TSWV), Pepino mosaic potexvirus (PepMV), Pepper mild mottle tobamovirus (PMMoV) ve Tomato brown rugose fruit tobamovirus (ToBRFV) and as a result 12% of the seeds were founded to be infected with TMV, 23% with ToMV and 8% with CMV. Within the scope of this study it was seen that some seed samples have mixed infections. 67 seeds of the 238 seeds has been found to be infected in this study. This is a rate of 28% were the seeds are infected at least with one viral agent.
