Özet
Bu çalışmada önemli tıbbi sülük türleri Hirudo verbana ve Hirudo sulukii’nin
vücut duvarında epidermis histolojisi ve salgı hücresi tiplerinin belirlenip çeşitli
histokimyasal boyamalarla salgı hücrelerinin boyanma özelliklerinin incelenmesi
amaçlanmıştır. Ayrıca epidermis epitel hücre alanları, salgı hücre tiplerinin alanları ve
salgı hücrelerinin histokimyasal boyalarla (Periodic Acid Schiff (PAS)) ve (Alcian Blue
(AB)) boyanan salgı alanları her iki türde istatistiksel olarak karşılaştırılmıştır. Basit tek
tabakalı silindirik epitel ve salgı hücrelerinden oluşan epidermisin üstünde epitel
hücrelerinin salgısı olan kütikula tabakası bulunur. Epidermise yakın bölgede Tip 1
olarak isimlendirilen salgı hücrelerinden bazıları armut şekilli salgı hücreleri olarak
belirlenmiştir. Vücut duvarının iç taraflarına doğru dermisten itibaren Tip 2a ve Tip 2b
hücreleri gruplar halinde yer almaktadır. Epidermise yakın salgı hücreleri PAS ve AB ile
zayıf boyanırken, Tip 2a daha koyu, Tip 2b salgı hücreleri ise en koyu hücreler olarak
belirlenmiştir. Çizgili kas tabakası halkasal, boyuna ve oblik yerleşim göstermektedir.
İstatistiksel veriler epitel ve salgı hücrelerin alanlarının genel olarak H. sulukii’de daha
büyük olduğunu göstermektedir. Bu durum H. sulukii’nin H. verbana’ya göre daha
dayanıklı bir tür olduğunu düşündürmektedir. Hem PAS hem AB boyamaları neticesinde
salgı hücrelerinin salgı alanının genel olarak H. sulukii’de daha fazla olması iki tür
arasında mukus içeriği bileşenlerinin farklı yoğunlukta olduğunu göstermektedir.
In this study, it was aimed to investigate the staining properties of secretory cells via histochemical stains and determine secretory cell types and epidermis histology in the body wall of important medicinal leeches called Hirudo verbana and Hirudo sulukii. In addition, areas of epidermis epithelial cells, areas of secretory cell types and secretory areas of secretory cells stained with histochemical stains (Periodic Acid Schiff(PAS) and Alcian Blue(AB)) were statistically compared in both species. The cuticle layer is located on the epidermis consisting of simple monolayer cylindrical epithelium and secretory cells. From some Type 1 cells located close to the epidermis were determined as pearshaped secretory cells. Type 2a and Type 2b cells are located in groups starting from the dermis towards the inner sides of the body wall. While the secretory cells close to the epidermis were stained weak with PAS and AB, Type 2a cells were darker and Type 2b secretory cells were the darkest cells. The striated muscle layer is located circular, longitudinally and obliquely. Statistical calculations showed that areas of epithelial and secretory cells are generally larger in H. sulukii than H. verbana. Therefore, it is considered that H. sulukii is more resistant species than H. verbana. Since both PAS and AB staining methods showed that secretory areas of secretory cells were larger in H. sulukii, we concluded that mucous composition between two species was different concentration.
