IV ÖZET BUĞDAYDA RAPD ( RANDOM AMPLIFIED POLYMORPHIC DNA ) MARKÖRLERİ İLE GENOTİPLERİN YORUMLANMASI ENGİN, Ayhan Yüksek Lisans Tezi, Tarla Bitkileri Bölümü Tez Yöneticisi: Prof. Dr. Süer YÜCE Nisan 1999, 50 Sayfa Bu çalışmada, 9 adet standart buğday varyetesi kullanılmıştır. Buğday tohumlan steril koşullar altında ekilmiştir. Buğdaylar 2-3 yapraklı olduğu zaman Doyle ve Doyle (1990) göre DNA izolasyonları yapılmıştır. Rastgele seçilen primerler aracılığıyla izole edilen DNA'lar % 1,5 agarose jelde yürütülmüştür. Jel Et Br ile boyanarak UV ışık altında incelenmiştir. Polymorphic ve monomorphıc bantlar raptanmıştır. Polymorfızm ve monomorfizm oranlan hesplanm ıştır. Genotiplerin moleküler düzeydeki farklılıklarının tespit edilmesinde RAPD yönteminin kullanılabilirliğinin tespiti ve genotiplerin DNA parmak izlerinin belirlenmesi yapılmıştır. V ABSTRACT TO COMMENT ON GENOTYPES OF WHEAT BY RAPD ( RANDOM AMPLIFIED POLYMORPHIC DNA ) MARKERS ENGİN, Ayhan MSc. Department of Field Crops Supervisor Prof.Dr.Süer YÜCE APRIL 1999, 50 pages In this study, nine standart weat varietes were used. The seed of wheat were planted under the sterile condition. Acording to Doyle and Doyle (1990) DNA isolations were made when seeds of wheat become 2 or 3 leaf. The isolated DNAs were amplified by using randomly selected primers. This amplified DNAs were run in the %1,5 agarose gel. The gel painting with EtBr were investigated under the UV light. Polymorphic and monomorphic bands were determined. Proportion of polymorphism and monomorphism were calculated. If RAPD method could be used for determining the differens in genotypes at moleculer levels were determined and to ascertain DNA fingerprints of genotypes were made.
