Özet
En önemli enfeksiyon hastalıklarından biri olan Leishmaniasis’in şiddetlenmesinden sorumlu tutulan Leishmania RNA virüsü (LRV), Yeni Dünya Leishmania türlerinde uzun süredir bilinmektedir ancak Eski Dünya türleri üzerinde kısıtlı sayıda araştırma bulunmaktadır. Bu çalışmada Türkiye’de görülen Leishmania türlerinde LRV varlığının araştırılması amaçlanmıştır. Viral çift-zincirli RNA (dsRNA), gDNA izolasyonu, RQ-DNAse uygulaması, RNA izolasyonu, cDNA eldesi ve uygun primerlerle hedef bölgenin çoğaltılması yöntemleri ile elde edilmiştir. Elde edilen sekans sonuçları daha önce bulunmuş olan LRV örnekleriyle karşılaştırılmıştır. İncelenen 29 örnekten (24 Leishmania tropica, 3 L. major ve 2 L. infantum) 10 tanesi (7 L. tropica, 3 L. major ) LRV varlığı açısından pozitif bulunmuştur. Yapılan BLAST analizi bu virüslerin LRV2-1 olduğunu göstermiştir. Bu çalışma ile dünyada ilk defa L. tropica türünde LRV saptanmıştır.
Leishmaniasis, one of the most important infectious diseases, has been known for many years in the New World Leishmania species, but there is limited research on the Old World species. In this study, it was aimed to investigate the presence of LRV in Leishmania species in Turkey. In order to obtain viral double-stranded RNA (dsRNA) , isolation of gDNA, RQ-DNAse application, RNA isolation, cDNA synthesis, amplification of target region with appropriate primers were applied. The sequence results were compared with the previous LRV specimens. Of the 29 samples tested (24 Leishmania tropica, 3 L. major and 2 L. infantum), 10 (7 L. tropica, 3 L. major) were found to be positive for LRV. BLAST analysis showed that these viruses were LRV2-1. With this study LRV was detected in L. tropica for the first time in the world.
