Öğe Cephalaria isaurica ve Cephalaria stellipilis türleri üzerindeki fitokimyasal çalışmalar(Ege Üniversitesi, 2010) Kayce, Peyker; Kırmızıgül, SüheylaÇalışmamızda Cephalaria isaurica ve Cephalaria stellipilis (Dipsacaceae) endemik bitkileri botanik olarak tanımlandıktan sonra fitokimyasal özellikleri ilk kez araştırıldı. Uygun koşullarda kurutulan ve öğütülen bitkilerin önce sırasıyla MeOH, MeOH:H2O, sonra elde edilen her bir ekstrenin n-Hegzan ve n-BuOH:H2O çözgen ve çözgen karışımlarıyla ekstraksiyonları yapıldı. Elde edilen ekstrelerin antioksidan (DPPH) ve antimikrobiyal (MIC) aktiviteleri test edildi. En aktif fraksiyon (n-BuOH) üzerinde yürütülen izolasyon ve saflandırma çalışmalarında çeşitli kromatografik (VLC, CC, MPLC, PLC, TLC) ve kimyasal (hidroliz, türevlendirme) yöntemlerden faydalanıldı. Saf olarak elde edilen bileşiklerin yapıları gelişmiş spektroskopik yöntemlerle (UV, FT-IR, 1D- ve 2DNMR, Kütle) belirlendi. Sonuç olarak, yapılan izolasyon, saflandırma ve yapı tayini çalışmalarında C. isaurica bitkisinden iki triterpen glikozit (bileşik 1 ve 2), bir C-flavon glikozit (bileşik 3), iki iridoit glikozit (bileşik 4 ve 5) ve bir iridoit aglikonu (bileşik 6) elde edildi. C.stellipilis bitkisinden ise, iki fenolik glikozit (bileşik 7 ve 8) ve bir C-flavon glikozit (bileşik 9) elde edildi. İlk bitkide tanımlanan iridoit aglikon (bileşik 6) ile bir iridoit glikozit (bileşik 4) bu bitkide de belirlendi. Elucidation processes of all pure compounds and biological properties of the new compounds were investigated on Cephalaria davisiana, Cephalaria aytachii and Cephalaria elazigensis var. purpurea (Dipsacaceae) plant species for the first time. The air dried and powdered plants collected and defined in flowering period were extracted with MeOH, MeOH: H2O, n-BuOH: H2O. Each n-BuOH extracts were extracted with n-Hexane for defeating oily parts. The isolation and purification studies on n-BuOH extracts were lead to use various chromatographic (VLC, CC, MPLC, TLC) and chemical (hydrolysis and derivatizations) methods. Structural determination of the pure compounds was carried out by modern spectroscopic (FT-IR, 1D-, 2D-NMR, HR-ESI/MS, ESI/MS and GC/MS) techniques. As a result, totally 27 natural products, 25 of which are glycosides, were obtained. Twelve glycosides from C. davisiana, 2 of which are new (Davisionaside A-B), 9 glycosides from C. aytachii, 1 of which is new (Aytachoside A), and 6 natural compounds from C. elazigensis var. purpurea, 1 of which is new (Cephoside A) and 1 of them (Anemoclemoside A) is detected on Dipsacaceae family for the first time, were investigated The antimicrobial and cytotoxic activity tests of the new compounds were evaluated using MIC and MTT assays, respectively.Öğe Description of biologically active natural products from three endemic cephalaria species(Ege Üniversitesi, 2016) Kayce, Peyker; Kırmızıgül, SüheylaBiyolojik aktif yeni doğal bileşiklerin elde edilmesinin amaçlandığı bu çalışmada, ülkemize endemik üç Cephalaria türü kimyasal ve biyolojik açıdan incelendi. Cephalaria davisiana, Cephalaria aytachii ve Cephalaria elazigensis var. purpurea bitki türlerinin doğal bileşik içeriği, tüm saf bileşiklerin yapılarının tayini ve yeni bileşiklerin biyolojik aktiviteleri ilk defa belirlendi. Çiçeklenme dönemlerinde toplanıp tanımlanan ve uygun koşullarda kurutulan bitkiler, öğütüldükten sonra sırasıyla MeOH, MeOH:H2O, n-BuOH:H2O ile ektre edildi. Elde edilen her bir n-BuOH ekstresinin n-hegzan ile ekstraksiyonu yapıldı. Glikozitlerce zengin n-BuOH fraksiyonları üzerinde yürütülen izolasyon ve saflandırma çalışmalarında çeşitli kromatografik (VLC, MPLC, CC, TLC) ve kimyasal (hidroliz, türevlendirme) yöntemler kullanıldı. Saf olarak elde edilen doğal bileşiklerin kimyasal yapıları gelişmiş spektroskopik yöntemlerle (FT-IR, 1D- ve 2D-NMR, ESI/MS, HR-ESI/MS ve GC/MS) belirlendi. Kimyasal ve spektroskopik işlemler sonrası 25 tanesi glikozit olmak üzere 27 doğal bileşik elde edildi. C. davisiana bitkisinden 2'si yeni (Davisionaside AB) 12 glikozit, C. aytachii bitkisinden 1'i yeni (Aytachoside A) 10 glikozit elde edildi. C. elazigensis var. purpurea bitkisinden de 1' yeni (Cephoside A), 1'i ise Dipsacaceae ailesinde ilk kez belirlenen (Anemoclemoside A) iki glikozit olmak üzere toplam 6 doğal bileşik elde edildi. Suleyman; Sumbul, Huseyin; Arda, Nazli; Kirmizigul, SuheylaThis paper focused on the assessment of fatty acid composition and antioxidant properties of ten Cephalaria (C. aytachii, C. taurica, C. tuteliana, C. procera, C. speciosa, C. tchihatchewii, C. hirsuta, C. elazigensis var. elazigensis, C. anatolica and C. aristata) species. The principal fatty acids in all species were oleic acid (10.28-31.65%), linoleic acid (17.81-37.67%) and palmitic acid (10.54-23.81%). Linolenic acid was also the most abundant fatty acid component in C. tuteliana (24.42%) and in C. speciosa (36.65%). In vitro antioxidant capacity of the hexane extracts of ten Cephalaria species was investigated by CUPRAC and DPPH methods. Total phenolic content of hexane extracts was also examined. The results showed that all species of Cephalaria have antioxidant properties with the highest trolox equivalent antioxidant capacity (1.005 +/- 0.13 mmol trolox equivalent per gram extact) in C. aristata and the highest radical scavenging activity (IC50 value 3.768 +/- 0.67 mg/mL) in C. tchihatchewii. It was found that reducing power of C. aristata and radical scavenging potential of C. tchihatchewii were mainly due to highest phenolic contents of these species (2.907 +/- 0.146 and 3.037 +/- 0.156 mg gallic acid equivalent per gram extract, respectively). These findings suggest that the Cephalaria species might be used as a potential source of unsaturated fatty acids as well as phenolic constituents possessing antioxidant activity in food, cosmetics and pharmaceutical industries.Öğe Isolation and characterization of biologically active glycosides from endemic Cephalaria species in Anatolia(Elsevier Science Bv, 2011) Sarikahya, Nazli Boke; Pekmez, Murat; Arda, Nazli; Kayce, Peyker; Yavasoglu, N. Ulku Karabay; Kirmizigul, SuheylaChemical constituents of the aerial parts of four endemic Cephalaria species (Dipsacaceae) in Anatolia, C. scoparia, C. paphlagonica, C. isaurica and C. stellipilis, were investigated. Two new triterpene glycosides from C. scoparia, namely scoposides F (1) and G (2), along with 11 known compounds including phenolic, flavonoid, iridoid and triterpene-type glycosides were isolated from these species. The structures of all compounds were elucidated on the basis of spectroscopic data and chemical evidence. Antioxidant activity of the compounds was tested with CUPRAC method for the first time in addition to the DPPH by using trolox and ascorbic acid as reference antioxidants, respectively. The antimicrobial activity was assessed by MIC assay and the results were given as MIC (mu g/mL) values. New triterpene glycosides scoposides F-G (1 and 2) and the compounds paphlagonoside A (6), paphlagonoside B (7) and isacoside (9), which were not tested previously, exhibited as strong antibacterial activity against Gram (+) and Gram ( ) bacteria. All rights reserved.Öğe Isolation and identification of a new saponin from Cephalaria aytachii(Taylor & Francis Ltd, 2017) Kayce, Peyker; Kirmizigul, SuheylaA new hederagenin-type triterpenoid glycoside (1) named Aytachoside A, along with eight known triterpene glycosides, were isolated from the aerial parts of Cephalaria aytachii Gokturk and Sumbul (Dipsacaceae). The structures of compounds 1-9 were determined by spectroscopic (1D and 2D NMR, HRESIMS) and chemical examinations. The antimicrobial effect of compound 1 was found considerably active against Escherichia coli, Pseudomonas aeruginosa and especially Salmonella typhimurium microorganisms using the MIC method. Compound 2, named anemoclemoside A, which is an unusual triterpene glycoside, was identified in the family Dipsacaceae for the first time. Chemical structures of all compounds were determined on the basis of the HRESIMS and 1D and 2D NMR data. The ethanol extracts from C. elazigensis var. purpurea, C. anatolica, and C. elmaliensis possessed the highest mortality against first instar Ae. aegypti larvae. Luteolin-7-O-beta-D-glycoside (1), isolated from C. elmaliensis ethanol extract, demonstrated 33% and 53% mortality at 0.1 mu g/mL concentration against first instar ORL (susceptible) and PR (pyrethroid resistant) strains, respectively. C. scoparia acetone extract showed 100% mortality against adult Ae. aegypti. From this extract compounds 2-8 were isolated. Compound 2 (isoorientin) possessed the highest toxicity with 31.7% and 65% mortality at a 10 mu g/mL concentration against adult ORL and PR strains, respectively. One new prosapogenin (1a) was also obtained after the alkaline hydrolysis of compound 1. The chemical structures of all compounds were established mainly by 1D-, 2D-NMR and HR-ESI/MS analysis as well as chemical methods. The antibacterial and antifungal effects of compounds 1-2 were evaluated against Gram-positive, Gram-negative bacteria and unicellular yeast C. albicans by MIC method.
