H. C.Öğe Composition of essential oil and fatty acids and antioxidant assays of Psephellus pyrrhoblepharus (Boiss.) Wagenitz(Innovhub Ssi-Area Ssog, 2019) Tastan, P.; Fafal, T.; Demirci, B.; Kivcak, B.In this study, the essential oil of Psephellus pyrrhoblepharus (Boiss.) Wagenitz was obtained by hydrodistillation using a Clevenger-type apparatus. the oil was analysed by GC and GC/MS system. the components were separated as for adherence to the column and evaluated as for relative rate. Mass spectra of each component were taken. GC and GC/MS analyses of the essential oil from P. pyrrhoblepharus determined the identification of 59 components representing 84% of the oil. With this analysis, the major component was found as hexadecanoic acid (17.1%). Subsequent to this component, other major components were spatulenol (10.3%), caryophyllene oxide (9.6%) and heptacosane (5.2%). in addition to this, fatty acid methyl esters (FAMEs) from P. pyhhoblepharus were prepared for analysis of fatty acids. By this test the amount of saturated fatty acids (SFA) were 31.37% with major fatty acid palmitic acid (12.86%). Also, the antioxidant assays DPPH, ABTS, ORAC, CUPRAC and reducing power assay were carried on methanol:water, n-hexane, chloroform and ethyl acetate extracts of P. pyrrhoblepharus. With the assays, ethyl acetate extract of P. pyrrhoblepharus was found as the most antioxidant extract. C.In this study, the fatty-acid profile, essential-oil composition and cytotoxic and antioxidant activities of the aerial parts of Ranunculus pedatus Waldst. & Kit. subsp. pedatus have been examined. Linoleic acid (24.47 %) and palmitoleic acid (18.94 %): were identified as major components by GC-FID and GC/MS analysis. Unsaturated fatty acids were found in higher amounts than saturated fatty acids. The essential-oil composition was also analyzed by GC and GC/MS and 29 components were characterized. Protoanemonin (14.3 %), phytol (13.7 %); alpha-bisabolol (7.9 %), (Z)-3-hexenol (7.8 %) and octanol (6.7 %) were determined as the main constituents of the essential oil. Ethanol and chloroform extracts were investigated for the cytotoxic activity against PC3, HeLa and Vero human cancer cell lines and both : of the extracts were found to be effective On HeLa. The IC50 values were determined as 47 and 78.4 mg/mL, for methanol and chloroform : extracts respectively. Antioxidant activity of chloroform and ethanol extracts were evaluated by DPPH assay. The total phenolics and flavonoid contents of the extracts were determined by Folin-Ciocalteau and aluminium chloride methods, respectively. The ethanol , extract was found to possess 67.54 % DPPH inhibition. The results obtained in the antioxidant activity assay were in positive correlation le with the total phenolics and flavonoid contents of the extracts.Öğe The essential oil composition of aerial parts of Anthemis tricolor Boiss. from Cyprus(Taylor & Francis Ltd, 2014) Karaalp, C.; Erel, S. (Asteraceae), an endemic taxon from Cyprus, was analysed by GC and GC-MS. The analysis revealed 40 compounds accounting for 84.5% of the oil. Muurola-4,11-diene (20.2%), beta-caryophyllene (9.1%) and germacrene-D (5.5%) were the main compounds of A. tricolor. Moreover, this is the first study on EO composition of endemicM. rotundifoliumBoiss. and chemical variabilities of the EOs of otherMarrubiumtaxa collected from Turkey.Öğe The influence of functional pinealectomy and exogenous melatonin application on healing of a burr hole in adult rat calvaria: a histological and immunohistochemical study(Via Medica, 2022) Basaloglu, H. O.Background: Even today, repair of the cranial defects still represents a significant challenge in neurosurgery and various options have been used for their reconstruction to date. However, there are very few studies investigating the effects of exogenous administration of melatonin (MEL) as an agent that promotes bone regeneration. The goal of this study was to investigate the effects of functional pinealectomy (Px) and exogenous MEL administration on the bone repair properties and surrounding connective tissue alterations in a rat calvaria model. Materials and methods: The total of 30 adult female Wistar-Albino rats was randomly divided into three groups (n = 10): control group (CO; 12 h light/12 h dark exposure), functional Px group (24 h light exposure, light-induced functional Px), and Px+MEL group (light-induced Px + MEL, 20 mg/kg/day for 12 weeks). Critical-sized burr-hole defects (diameter: 3.0 mm) were surgically created by a single operator in the calvarium of all rats, using an electric drill. Animals in Px+MEL group received MEL 20 mg/kg/day for 12 weeks. At the end of the study, bone healing and connective tissue alterations surrounding drilled defect area in the rat calvaria were determined in haematoxylin-eosin-stained and Mallory Azan slices applied in anti-bone sialoprotein. Image Pro Express 4.5 programme was used for histomorphometric calculation of areas of new bone and fibrotic tissue. Normality control was performed by Shapiro-Wilk test. Variance homogeneities were examined by Shapiro-Wilk and Levene tests; Tukey HSD test was used as a post hoc method since there was no homogeneity problem. All hypothesis tests were performed at the 0.05 significance level. Results: Histological analysis showed that the bone repair process in the Px+MEL group was similar to that of the CO group, whereas the functional Px group showed a delay. Histomorphometrically, it was found that the Px group had the largest hole diameter and the most fibrotic scar area, although no binary statis- tical significance was found between the CO and Px+MEL groups (p = 0.910).In terms of vascularisation, it was observed that the most vascular structure was found in the Px+MEL group among the scar tissue and ossification areas, while the vascularisation was the least in the Px group (p < 0.001). Conclusions: Our findings revealed that bone repair process was impaired in functional Px group, but exogenous MEL replacement was able to restore this response. Thus, it is concluded that utilisation of MEL may improve the bone repair in calvarial defects. (Folia Morphol 2022; 81, 2: 271-279)
