Öğe Effect of Different Media on Short Term Storage of Leaping Mullet (Liza saliens (Risso, 1810)) Sperm(2020) Engin, Serhat; Fırat, M. Kürşat; Ozden, Osman; Suzer, Cüneyt; Hekimoğlu, Müge; Antepli, Onurkan; Saka, ŞahinThe present study aimed to evaluate the activation characteristics of the leaping mullet spermatozoa and diluted with four different media (D1, D2, D3, D4) for 250 h at 4±2°C. The semen was collected from nine wild fish in Homa Lagoon, İzmir, Turkey in July, during reproductive season. In each trial (n = 9), semen samples were collected and mixed to reduce sperm quality variation to obtain the required gametes for the experiment. The semen was activated with natural seawater (salinity of 34, pH 7.9 and 4±2°C) to determine: motility time, motility rate and viability of sperm. Dilution with inactivation media 1:3 increased motility time of diluted semen than control group. At the end of the experiment, no significant difference was found for the duration of survival in the D2 and D4 groups. In this context, Bacillus was supplemented via rotifer feeding and water and its effects on pancreatic and intestinal enzyme activities as well as aquaculture parameters were evaluated during early life development. In the experimental group, as probiotic three Bacillus sp. spores were introduced via rotifer and larval culture tanks, while the larvae in control group did not feed any probiotic supplementation. At the end of the experiment on 40 days after hatching, the probiotic-supplemented group exhibited better growth performance and there were statistically differences in between groups of probiotic-treated and control regarding growth parameters (P < 0.01), despite insignificant survival rate (P > 0.05). In terms of enzymatic expressions, S. aurata larvae receiving probiotic supplementation through rotifers demonstrated noteworthy (P < 0.05) enhancements in specific activities of pancreatic and intestinal enzymes, except for amylase (P > 0.05), when compared to the control group. It is concluded that the administration of Bacillus sp. as probiotic bacteria through rotifer supplementation and water intake demonstrates significant positive impacts on both growth parameters and specific activities of main pancreatic and intestinal enzymes of seabream larvae.Öğe Topan kefal (Mugil cephalus) spermlerinin farklı ortamlarda kısa süreli korunumu(Ege Üniversitesi, Fen Bilimleri Enstitüsü, 2018) Antepli, Onurkan; Saka, ŞahinBu araştırmada, Ege Üniversitesine bağlı Homa Dalyanından topan kefaller hasat edilmiştir. Temin edilen sperm örnekleri 0-2 °C'deki buz kalıpları içinde muhafaza edilmiştir. Daha sonra her saatte bir faz kontrast tip mikroskopta incelenmiştir. Bu çalışmada, topan kefal (Mugil cephalus L. 1758) spermasının doğal üreme mevsimi süresince pH, hacim, yaşama kabiliyeti ve muhafaza süresi gibi spermatolojik özelliklerin belirlenmiştir. Ayrıca kısa süreli koruma işleminde 4 farklı inaktivatörün (Hepes, Tris, BSA, DMSO) etki mekazimalarını saptanmıştır. Çalışmada, toplam 5 adet anaç balık sperma alımı için kullanılmıştır. Kullanılan anaçların ortalama ağırlıkları 753,6±47,21 g, ortalama boyları 40,6±1,14 cm olarak saptanmıştır. Çalışmada kullanılan topan kefal spermin pH değeri 6,7 olarak saptanmıştır. Anaçlardan elde edilen spermlerin ortalama hacmi 2±0,5 ml.kg-1 olarak tespit edilmiştir. Spermlerin yaşama kabiliyeti taze sperma da 6 dk, hepes de 6 dk 30 sn, bsa da 5 dk 20 sn, tris de 3 dk 20 sn ve dmso da 8 dk 30 sn olarak tespit edilmiştir. 36 saatlik çalışma periyodu sonunda, spermlerin muhafaza süreleri taze sperma da 28 saat, inaktivatörlerden tris de 2 saat, diğer inaktivatörlerde (Hepes, BSA ve DMSO) 36 saat hareketlilik tespit edilmiştir.
