ÖZET ERKEN YAŞTA MİYOKARD İNFARKTÜSÜ GELİŞEN OLGULARDA FAKTÖR XIII (FXIII) VAL34LEU GEN POLİMORFİZMİNİN KATKISININ ARAŞTIRILMASI HANÇER, Veysel Sabri Yüksek Lisans Tezi, Biyoloji Bölümü Tez Yöneticisi: Prof. Dr. Gürsel ERGEN Eylül 2002, 61 sayfa Bu çalışmada oluşumunun çok etkenli olduğu miyokard infarktüsü'ne, Faktör XIII (FXIII) Val34Leu gen polimorfizminin katkısının olup olmadığı incelenmiştir. Çalışmaya miyokard infarktüs tanısı konmuş 117 genç hasta ve 126 sağlıklı kontrol dahil edilmiştir. FXIII Val34Leu polimorfizmi, periferik venöz kandan elde edilen DNA'lar uygun primerler ile 'Polimeraz Zincir Reaksiyonu (PCR), tekniği' kullanılarak çoğaltılmıştır. Hin 61 enzimi ile PCR ürünleri kesilip agaroz jelde görüntülenmiştir. Böylece heterozigot ve homozigot bireyler belirlenmiştir. Hasta ve kontrol grubunun sonuçlan istatistiksel metodlarla karşılaştırılmıştır. Sonuçta hasta grubunda 93 bireyde W, 24 bireyde VL genotipi saptanmıştır (V allel sıklığı %89.7, L allel sıklığı % 10.2'dir). Kontrol grubunda 79 bireyde W, 45 bireyde VL, 2 bireyde LL genotipi saptanmıştır (V allel sıklığı %80.5, L allel sıklığı % 19.4'dir) aradaki fark istatistiksel olarak anlamlı bulunmuştur (SPSS Ver. 10.0 programında p = 0.002). Bu bulgular, FXIII geninde VL polimorfizminin miyokard infarktüsünden koruyucu bir etkiye sahip olduğunu desteklemektedir. vn ABSTRACT A STUDY ABOUT THE EFFECT OF FACTOR XHI (FXm) VAL34LEU GENE POLYMORPfflSM IN MYOCARDIAL INFARCTION AT EARLY AGES HANÇER, Veysel Sabri MSc in Biology Supervisor: Prof. Dr. Gürsel ERGEN September 2002, 61 pages In this study, whether factor XIII Val34Leu polymorphism has any effect on the evolution of myocardial infarction which is known to be the result of multiple etiological factors have been evaluated. 117 patients with myocardial infarction and 126 healthy controls were included in the study. To detect FXHI Val34Leu polymorphism, DNA extracted from venous blood leucocytes were replicated and analyzed by 'PCR method' using spesific probes. PCR products were digested by Hin6I enzyme and visualized by agarose gel electrophoresis. So bom homozygous and heterozygous cases were detected. The results of patients and control group were compared by statistical methods. 93 patients were found to have W genotype (V allele incidence was 89.7 %, and L allele incidence was 10.2 %). In the control group 79 showed W, 45 showed VL and 2 showed LL genotype (V allele incidence was 80.5 % and L allele incidence was 19.4 %). The difference was statistically significant (SPSS Ver.l0.0p=0.002). These findings support the hypothesis that VL polymorphism in the FXHI gene has a protective effect against myocardial infarction.
