Altı membran gec?is?li prostat proteini 2 / six transmemrane protein of prostate 2 (STAMP2) geni, prostat kanserinde (PCa) androjen tarafından du?zenlenen bir gendir. STAMP2 ektopik ekspresyonu, hu?cre c?og?almasını ve kolonizasyonunu arttırdıg?ı bulunmus?tur. Farklı hu?cre hatlarında yapılan c?alıs?malarda ve protein yapısında belirlenen bo?lgelerde, hu?cresel metabolizmada bir rolu? olabileceg?i du?s?u?nu?lmektedir. STAMP2'nin bu o?nemli go?revlerini anlayabilmek ic?in; PCa hu?cre hatlarında, farklı metabolik kos?ullarda, SIRT1 proteini ile olan ilis?kisi ve bu regu?lasyonun hu?creye etkilerinin aras?tırılması amac?lanmıs?tır. Bu tez c?alıs?masında STAMP2 ve SIRT1 ekspresyon durumlarının antagonist bir regu?lasyonda oldug?u ve bu durumun hu?cre fenotipine etki ettig?i go?zlenmis?tir. Bu regu?lasyon PCa hu?crelerinin c?og?alması ve sag?kalım mekanizmalarını daha iyi anlamamızı sag?layarak, terapo?tik tedavilerin etkisini arttıracak o?neme sahiptir. Sonuc?larımız, farklı metabolik kos?ullarda STAMP2 ile SIRT1 regu?lasyonunun mitotik aktivite, oksidatif stress yanıtı ve DNA hasar onarımı sinyal yolaklarında yer alan proteinleri etkiledig?i ve yapılan analizlerde bu parametrelerin hu?cre c?og?almasını du?zenledig?i go?sterilmis?tir. The six transmembrane protein of prostate 2 (STAMP2) is an androgen- regulated gene whose mRNA expression is increased in prostate cancer (PCa). Ectopic expression of STAMP2 in prostate cancer cells significantly increases cell proliferation and colony formation. Metabolic studies in different cell lines and identified domains in the protein structure, these data established that STAMP2 may play a role in cellular metabolism. To be able to elucidate the function of STAMP2, we aimed to investigate the relationship between SIRT1 protein and cellular effects of this regulation on PCa cell lines under different metabolic conditions. In this study, we showed that STAMP2 and SIRT1 expression states are in an antagonist regulation and this affects the cell phenotype. The regulation allows us to better understand the proliferation and survival mechanisms of PCa cells, which is important to increase the effect of therapeutic treatments. Further, we found that, under different metabolic conditions, regulation of STAMP2 and SIRT1 has affected mitotic activity, oxidative stress response and DNA damage repair signaling pathways proteins and signal pathways proteins. In the analyzes performed, it has been shown that these parameters regulate cell proliferation. Keywords: STAMP2, prostate cancer, SIRT1, AR signaling pathways.
