Düzce ve Adapazarı'nda yetiştirilen yabancı tütün çeşitlerinde görülen virutik belirtilerin etmenlerinin belirlenmesi üzerinde araştırmalar

Özet
Ill ÖZET Bu çalışma, 1995 yılı üretim mevsiminde Düzce ve Adapazarı yörelerinde Virginia ve Burley tütünlerinin yetiştirildiği alanlarda virusia bulaşık olduğundan şüphe edilen tütün bitkilerinden alınan yaprak örnekleri incelenerek gerçekleştirilmiştir. Konuyla ilgili tüm denemeler, Ege Üniversitesi Ziraat Fakültesi Bitki Koruma Bölümü'nde yürütülmüştür. Yalnız Patates X Virusu ile Patates Y Virüsünün tanılanması için yapılan ELISA testleri Menemen Tarımsal Araştırma Enstitüsü'nde uygulanmıştır. Bazı test bitkilerine mekanik inokulasyon yapılarak ve DAS-ELISA (Double Antibody Sandwich Enzyme-Linked Immunosorbent Assay) yöntemi kullanılarak, Burley ve Virginia tütünlerinden alınan yaprak örneklerindeki viruslar tanılanmaya çalışılmıştır. Çalışmada elde edilen sonuçlar şu şekilde özetlenebilir: 1. Virginia ve Burley tütün bitkilerinden alınan yaprak örnekleri kullanılarak test bitkilerine yapılan mekanik inokulasyon sonucunda; mozayık, kabarcıklı mozayık, yaprak deformasyonu, nekrotik veya klorotik lokal lezyon, renk açılması, damarlarda bantlaşma, tezyinatlı mozayık gibi virus belirtileri meydana gelmiştir. 2. Serolojik olarak testlemede ise Tütün Mozayık Virusu, Hıyar Mozayık Virusu, Patates X Virusu, Patates Y Virusu ve Yonca Mozayık Virusu tanılama kitleri kullanılarak DAS-ELISA yöntemi uygulanmıştır. Örneklerin büyük bir çoğunluğunda Hıyar Mozayık Virusu, bir kısmında da Tütün Mozayık Virusu saptanmıştır. Bunun yanı sıra her iki virüsün karışık enfeksiyonları da gözlenmiştir. Patates X Virusu, Patates Y Virusu ve Yonca Mozayık Virusu için yapılan ELISA testinde ise herhangi bir pozitif sonuç elde edilememiştir
ABSTRACT This survey was performed in the growing season of 1995 and the leaf samples were obtained from the Burley and the Virginia tobacco plants showing as virus symptom's in Düzce and Adapazarı districts. Except of ELISA test method for both Potato X Virus and Potato Y Virus, the studies were carried out in Ege University Agriculture Faculty Department of Plant Protection. Only the ELISA test method of Potato X Virus and Potato Y Virus, was carried out in Menemen Agricultural Research Institute. The viruses in the Burley and the Virginia leaf samples collected were determined considering the symptoms on test plants and the results from DAS- ELISA (Double Antibody Sandwich Enzyme-Linked Immunosorbent Assay) method. The findings from this study can be summarized as follows:. 1. When the leaf samples collected from the Virginia and the Burley tobacco plants were inoculated to certain test plants and they showed the typical virus symptoms such as mosaic, blister mosaic, leaf deformation, necrotic or chlorotic local lesion, yellowing, vein banding, mottle mosaic. 2. In serologic property was used DAS-ELI SA method, by using the ELISA kits of Tobacco Mosaic Virus, Cucumber Mosaic Virus, Potato Y Virus, Potato X Virus and Alfalfa Mosaic Virus. It was found that, the majority of the samples was infected with Cucumber Mosaic Virus. Moreover, in remaining samples Tobacco Mosaic Virus was detected. On the other hand, some samples contained the two viruses together as mixed infections. Potato X Virus, Potato Y Virus and Alfalfa Mosaic Virus were not found in the leaf samples obtained.
