Özet
Küçük hücreli akciğer kanserinde (KHAK) mikrosatellit instabilite nadir görülmektedir. Heterozigotluğun kaybı (LOH) ise 1p, 3p, 5q, 8p, 9p, l Öp, 1 ip, 13q, 22q ve 17 kromozom lokalizasyonlanndaki bazı dinükleotid tekrarlarda belli oranlarda bulunmuştur. Araştırmamızda 5q 13. lokusunda bir instabilite, LOH ya da çalışılan dinükleotid tekrarın (D5S679) popülasyona spesifik bir allel gidişinin varlığı araştırılmıştır. Çalışmamızda analiz edilen 27 örneğin 25'inde (%92,6) D5S679'un 6 nolu alellinin var olduğu görülmüştür. Bu alel 39 normal kontrol grubunda sadece %2.6 oranında gözlenmiştir (p<0.0001). 27 KHAK örneğinin 6'sında hastaların normal akciğer dokusuna ait örnekler çalışılıp karşılaştırıldığında 4'ünde LOH, ikisinde mikrosatellit instabilite gözlenmiştir. KHAK'de 5S679 dinükleotid tekrarla yapılan herhangi bir çalışma ise bildirilmemiştir. Altı nolu alelin bu yüksek oranı KHAK'de D5S679'un önemli bir dinükleotid tekrar olduğunu ve Türk popülasyonunda KHAK ile birlikte giden spesifik bir alel bulundurduğunu göstermiştir. Aynı zamanda bu mikrosatellit, mikrosatellit instabilite ve LOH çalışmaları için uygun bir para-metredir.
Microsatellite instability has been rarely reported in small-cell lung cancer (SCLC). Meanwhile, loss of heterozygosity (LOH) was found in certain amounts on some dinucleotide repeats situated on chromosomal locations 1p 3p, 5q, 8p, 9p 10p, 1ip 13q, 22q and 17. We studied the presence of specific dinucleotide repeat (D5S679) on 5q 3 for instability, the presence of LOH and diverse alleles specific to our population as well as their frequency. The allele number six of D5S679 was found in 25 of 27 (92.6%) cases analyzed in our study. However, the same allele was encountered only in two of 39 (2.6%) individuals in normal control group (p<0.0001). When tumor tissue was compared to non-tumor lung tissue in only six cases among 27 cases with SCLC, LOH was found in four and microsatellite instability in two. A study of D5S679 in SCLC was not reported previously. The high frequency of the allele 6 of D5S679 in SCLC shows that it could be a significant marker for this disease and it contains a specific allel within SCLC in Turkish population. In conclusion, this microsatellite is an appropriate parameter for MI and LOH studies.
