Özet
En geniş tanımıyla ilaç geliştirilmesi kavramı, bir hastalığın veya terapötik hedefin
tespiti ile başlayan, tasarlanan bileşiklerin sentezi, hayvanlarda farmakolojik
farmakokinetik ve güvenlik çalışmalarıyla devam eden ve insanlarda faz I ve faz II klinik
çalışmalarıyla tamamlanan bir süreçtir. Dünya Sağlık Teşkilatının 1985 yılında
oluşturduğu temel ilaçlar listesinde bulunan 252 ilaçtan 123 tanesi (% 48.9) kimyasal
sentez yoluyla elde edilmektedir. Yeni ilaçların geliştirilmesi amacıyla her yıl bir çok yeni
bileşik devamlı olarak sentez edilerek, test edilmektedir.
Departmanımızda devam eden çalışmalar, yeni ilaçların ya da yeni lider
moleküllerin geliştirilmesi çalışmalarını da kapsamaktadır. Bu çabaların bir parçası olarak,
bu çalışmada konjuge karbonilen türevleri ve heterosiklik halka taşıyan bileşikler üzerinde
sentez ve kimyasal reaktivite çalışmalarının gerçekleştirilmesi amaçlanmıştır. Elde edilen
bileşiklerin sentezi ve spektral yapı teyitleri rapor edilmektedir.
The broadest definition of drug discovery is one defines as a process that starts with the identification of a disease and therapeutic target of interest and includes synthesis of the compounds designed and pharmacological, pharmacokinetic and safety studies in animals, followed by Phase I and Phase II clinical studies in humans. Of the 252 drugs included in the model list of essential drugs of World Health Organization of 1985 , 123 (48.9%) are obtained by chemical synthesis. Several new compounds are being contiually synthesized and tested every year to develop new drugs. The continuing studies in our department also includes the development of new drugs or new leads. As a part of those attempts, the synthesis and chemical reactivity studies on conjugated carbonylene derivatives and compounds bearing heterocyclic ring and have been aimed in this study. The synthesis and spectral confirmations of the compounds obtained are reported.
