Inherent non-linearities in immobilized enzyme assays and possibilities for selectivity in enzyme immobilization

Özet
Specific activities of immobilized ?-amylases toward native amylose and soluble starch have been determined at various immobilized enzyme concentrations. The resultant 'activity versus enzyme concentration' curves are shown to display inherently non-linear behaviour arising from the polymeric character of the substrate. Based on the conclusions drawn from the nature of the non-linearities it is hypothesized that the enzyme molecules tend to concentrate more on the surface of the carrier than inside the pores as the reactivities of the components in the coupling reaction are increased. © 1983 Science and Technology Letters.
