Özet
25 ÖZET Çalışma materyali olarak gelişen (-11 günlük) Albino fare (Mus muBCulus) embriyoları kullanılmıştır. AB (Alcian-Blue 8 GX) boyama yöntemiyle ile pH 2.5 ve 1 ile, pH 5.8 de 0.025 M, z. 0.06 M, 0.3 M, 0.65 M ve 0.9 M MgCl2 iyon konsantrasyonunda it 3 GAG (Glikozaminoglikan) ' ların dağılışı ışık mikroskobuyla incelenmiştir. Genel olarak pH 2, 5 ' ta yapılan boyamalarda GAG 'lar daha koyu canlı bir boyanma özelliği göstermiştir. pH 5. 8 ' de 0.025 M ve 0.06 M MgCİ2 iyon konsantrasyonunda yapılan boyamada pH 2. 5 ' la aynı özelliğe sahiptir pH l'deki boyamalar biraz daha soluk renklidir. pH 5, 8' de 0.3 M MgCl2 iyon konsantrasyonu da pH l'e benzerlik göstermektedir. 0.65 M ve 0.9 M MgCİ2 iyon konsantrasyonunda ise mavi renk belirgin değildir. AB boyama sonucunda göz bölgesi hariç ektodermal or j inli beyin, omurilik ve burun bölgesinde ağırlıklı olarak Hiyalüronik Asid, Kondroitin Sülfat ve Dermatan Sülfat, az miktarda ise Heparin, Heparan Sülfat ve Keratan Sülfat gözlenmiştir. Göz bölgesinde ise Ch-S, HA ve DS bulunmuştur. Endodermal or j inli özofagus, ana bronş ve bronşlar ile mezodermal orjinli damarlarda Hiyalüronik Asid, Kondroitin Sülfat ve Dermatan Sülfat vardır. Mezenşim hücreleri etrafında ve mezenşimden farklılaşan kıkırdak dokusunda da yalnızca Hiyalüronik Asid, Kondroitin Sülfat ve Dermatan Sülfat 'a rastlanmıştır
26 SUMMARY The material used in this work embryos, Mus musculus, of -11 days age. Distributions of GAGs stained in AB at pH 2,5 and 1 without MgCl2 and at pH 5.8 with different MgCl2 ion concentrations (respectively 0.025 M, 0.06 M, 0.3 M, 0.65 M and 0.9 M) were observed with light microscope. In generally, GAGs stained at pH 2.5 -showed darker and brillant staining characteristics. Similarly GAGs which were also stained at pH 5.8 with 0.025 M or 0.06 M MgCl2 ion concentrations showed the same staining characteristics with that ones stained at pH 2.5. However, GAGs which were stained at pH 1 were lighter and not as dark as the others mentioned above. GAGs which were stained at pH 5.8 with 0.3 M MgCl2 ion concentrations had the same staining properties with that ones stained at pH 1. The blue color of the GAGs stained at pH 5.8 with 0.65 M and 0.9 M MgCl2 ion concentrations were lighter than the others. As the result of AB staining, in the regions originated from the ectoderm such as brain, spinal cord and nose, except the region of eye, it was observed that HA (Hyaluronic Acid), Ch-S (Chondroithin sulfate) and DS (Dermatan Sulfate) were in high amount whilst HP (Heparin), HS (Heparan Sulfate) and KS (Keratan Sulfate) were vaguely. However, in the region of eye only Ch-S, HA and DS was observed. In the Oesophagus, bronchus and bronchiolles originated from endoderm and in the vascular structure originated from mesoderm only HA, Ch-S and DS were found. In all around of the mesenchimal cells and in the cartilage tissue differentiated from the mesenchim only HA, Ch-S and DS were found.
