Özet
İkinci nesil antiepileptik ilaçlardan biri olan Zonisamid (ZNS), diğer antiepileptik ilaçların mekanizmasından tamamen farklı ve kimyasal olarak sülfonamid olarak sınıflandırılan geniş spektrumlu bir ilaçtır. ZNS'nin güvenliği, etkinliği ve kontrolü klinik çalışmalarda geniş çapta kanıtlanmış, diğer antiepileptiklere göre ilaç-ilaç etkileşim potansiyeli düşük bir ilaç olduğu bildirilmiş ve nöbet kontrolünde bir tedavi seçeneği olarak önerilmiştir. Yapılan çalışmada 68Ga ile ZNS'in radyoişaretleme potansiyelinin belirlenmesi amaçlanmıştır. Bu amaç doğrultusunda radyoişaretleme, kalite kontrol ve stabilite çalışmaları yapılmış ve kromatografik yöntemlerden faydalanılmıştır. ZNS'nin 68Ga ile doğrudan işaretlenmesi sonucunda % 95 ± 4,24 radyoişaretlenme verimi elde edilmiştir. Sonuç olarak epileptik süreçler üzerindeki etki mekanizmaları düşünüldüğünde, anti-epileptik ilaçları görüntüleme özelliklerine sahip bir radyo izleyici ile birleştirerek; epilepsi gibi nörolojik hastalıklara etki edebilen görüntüleme radyoligandlarının sentezi üzerine çalışmalar yapılmıştır. Radyoişaretli antiepileptik ilacın nörolojik süreçlerin belirlenmesindeki potansiyelini araştırmak amacıyla daha ileri çalışmalar planlanacaktır. Zonisamide (ZNS), one of the second-generation antiepileptic drugs, is a broad-spectrum drug that is completely different from the mechanism of other antiepileptic drugs and is chemically classified as a sulfonamide. The safety, efficacy, and control of ZNS have been widely proven in clinical trials, it has been reported to be a drug with a low drug-drug interaction potential compared to other antiepileptics, and it has been recommended as a treatment option in seizure control. The aim of the study was to determine the radiolabeling potential of ZNS with 68Ga. For this purpose, radiolabeling, quality control and stability studies were carried out and chromatographic methods were used. As a result of direct labeling of ZNS with 68Ga, a radiolabeling efficiency of 95 ± 4.24% was obtained. As a result, considering the mechanisms of action on epileptic processes, by combining anti-epileptic drugs with a radio tracer with imaging properties; Studies have been conducted on the synthesis of imaging radioligands that can affect neurological diseases such as epilepsy. Further studies will be planned to investigate the potential of a radiolabeled antiepileptic drug in the determination of neurological processes.
