Biyoteknik yöntemle bitki korumada besin cezbedici formülasyon geliştirilmesine yönelik analitik yaklaşımlar

Özet
Son yıllarda gıda temini ve tarımsal üretimin sürdürülebilirliği kritik önem kazandığından, organik tarım uygulamaları ön plana çıkmıştır. Akdeniz meyvesineği (AMS) meyveye verdiği zarar ile kalitesini etkilediği gibi, ticari değerini de düşürmektedir. Önemli bir meyve ihracatçısı olan ülkemizde bu zararlı ile mücadelede alternatif yöntemlerin ortaya konması gereği doğmuştur. Bu tez çalışmasında AMS zararlısına karşı biyoteknik yöntemle mücadele amacıyla besin cezbedicisi formülasyon geliştirilmesi üzerinde durulmuştur. Karışımların etkinlikleri Y tüp olfaktometre düzeneği ile test edilmiş ve Plackett-Burman deney tasarımı ile öne çıkan etkin kimyasallar belirlenmiş ardından, bu kimyasalların optimum düzeyleri Merkezi Kompozit Tasarım ile saptanmıştır. İki adet aday formülasyon kontrollü salımın gerçekleşeceği tekstil malzemesine emdirilmiş, aday ambalaja yerleştirilmiş ve ruhsatlı ürün ile karşılaştırmalı olarak BZMAE bünyesindeki meyve bahçelerinde uzun süreli saha denemeleri gerçekleştirilmiştir. İki aday ürünün AMS yakalama performansları değerlendirildiğinde; bir tanesinin ruhsatlı ürüne yakın performans gösterdiği, diğerinin ise ruhsatlı üründen daha yüksek performans gösterdiği görülmüştür. Bu formülasyonların uçucu bileşenlerinin salım hızını ticari ürünle kıyaslamak için gerek bahçe denemelerinde gerekse dış ortam koşullarından etkilenmeden farkı sıcaklıklarda tutulan iklim odalarında bekletilen numunelerin hafta bazında kütle kaybı ve pH ölçümleri ve iklim odaları için ise majör ve minör bileşenlerin kromatografik analizleri gerçekleştirilmiştir.
Since the sustainability of agricultural production and food supply have gained critical importance in recent years, organic farming practices have become more important. The Mediterranean fruit fly (AMS), not only affects the quality of the fruit with its damage, but also reduces its commercial value. In our country, which is an important fruit exporter, it has become necessary to introduce alternative methods in the fight against this pest. In this thesis, the development of a food attractant formulation fighting against the AMS pest, with the biotechnical method was emphasized. The efficacy of the mixtures was tested with a Y tube olfactometer, and the prominent active chemicals were determined with the Plackett-Burman experimental design, and then the optimum levels were determined with the Central Composite Design. Two candidate formulations were impregnated with the textile material where controlled release would take place, placed in the candidate package and long-term field trials were carried out in the orchards of BZMAE in comparison with the licensed product of the candidate products. When the AMS catching performances of the products were evaluated; it was observed that one of them performed close to the licensed product, while the other one showed higher performance than the licensed product. In order to compare the release rate of the volatile components of the developed formulations with the commercial product, mass loss and pH measurements on a weekly basis of the samples retained in garden trials and climate chambers kept at different temperatures without being affected by outdoor conditions and, chromatographic analyzes of major and minor components for the samples from climate chambers were carried out.
