Özet
Yapay tohumlama, hayvan ıslahında kullanılan ilk biyoteknolojik
yöntemdir. 20. YY.‟dan itibaren hayvancılıkta yoğun olarak kullanılmaya
başlanmış, hayvancılık ıslah programlarının vazgeçilmez bir parçası olmuş,
hayvan ıslahına büyük ivme kazandırmıştır.
Hayvancılıkta yapay tohumlama yöntemlerinin yoğun kullanımıyla birlikte,
tohumlamanın ilk ve en önemli aşaması olan “Sperma alma, muayene ve
değerlendirme yöntemleri” de zaman içinde gelişmiştir. Makroskopik muayene ve
değerlendirme yöntemleri, daha sonra sperma değerlendirmeye yönelik özel
mikroskopların geliştirilmesiyle mikroskopik değerlendirme yöntemleri
geliştirilmiştir. Son yıllarda Bilgisayar teknoloji ve programlama yöntemleri ile
görüntüleme tekniklerinin gelişmesiyle birlikte daha hızlı, daha doğru muayene ve
değerlendirme yöntemleri geliştirilmiş ve bu yöntemler de genel olarak
“Bilgisayar Destekli Sperma Analiz Yöntemleri” (Computer Assisted Semen
Analysis CASA) olarak adlandırılmıştır.
Bu çalışmada klasik ve çağdaş sperma analiz yöntemleri tanıtılmış, CASA
yöntemlerinin getirdiği avantajlar vurgulanmış ve örnek bir çalışma olarak E. Ü.
Z. F. Küçükbaş Hayvan Biriminde mevcut olan “Spermvision” CASA sisteminin
bilimsel çalışmalarda kullanılmasını kolaylaştırmak amacıyla cihazın kullanım
kılavuzunun ayrıntılı açıklaması yapılmıştır.
The artificial insemination (A.I.) is the first biotechnological method used in the animal breeding. This methode were used very intensively in the Animal Science since at the begining of the 20th century. Therefore the amimal breeding programs would be faster realised and no animal breedings program could be planned without A.I. Programs. The semen analysing methods are devoloped also in the same period. With intensive using of artificial insemination methods in the livestock, the first and most important step of insemination like semen analysing including semen collection, semen examination and evaluation methods are , it has developed over time. The traditional methods of semen analysing like macroscopic and microscobic examination and evaluation methods are developed also. At the same time it has been observed very increasing improvements in the computer technology and programming methods and imaging techniques. In the last years were these improving techniques combined and developed new semen analysing methodes like CASA systems. Thus the semen analyising would be better, faster and easier and obtained faster and more trust results. These methods are generally called CASA. In this study, the classical and new modern techniques of semen analysis methods were introduced and compared. The advantages of the Casa methods were explained and as a sample methode “Spermvision™” was described. The user‟s manual and using principles of Spermvision™ system in the Semen Laboratory of E.U.Z.F small ruminants unit were explained detailed
